Schematic strategy for whole-mtDNA-genome PCR amplification & sequencing

Red boxes are short L-strand (5'3') PCR primers; blue boxes are primers for the reverse complement H-strand. Sixteen such primer pairs amplify the entire mtDNA molecule as an ordered series of sixteen overlapping ~1Kbp contigs (longer red- & blue-tipped boxes). Forward- and reverse-sequencing of all products in the set produces the complete mtDNA genome sequence for an individual.

The strategy differs substantially from conventional genomic sequencing based on random ("shotgun") sequence assembly
