        For each of the following questions, write a 500 ~ 750 word essay. The essay should show your understanding of the lecture material, demonstrate that you have prepared the answers ahead of time, and wherever possible the use of outside materials.

        For the exam, I will select FOUR of the questions, and you must answer any THREE. I sometimes require that one question in particular be answered, and then you have your choice of any TWO of the others.

         I want you answer all questions, in detail, as part of the learning experience. The Exam then confirms that you did the assignment.


1.    Distinguish between Genetics, Molecular Biology, and Biotechnology. Illustrate the differences by citing one example of each, and showing the degree to which the example fits best into one type of science, and not so much the others.

2.    What is a "Gene"? Compare and contrast notions of the eukaryotic gene after "cracking" of the Genetic Code in 1965 with contemporary notions in 2024.

I have described DNA replication as “DNA xeroxing”, and RNA transcription from DNA as making a “Silly Putty” copy. In what ways are these descriptions accurate? In what ways are these descriptions inaccurate?

4.    Explain how different kinds of SNP variation may alter the DNA sequences of genes, and the consequences for organisms.

5.    Describe the process of obtaining a DNA sequence of a particular gene, with emphasis on the biochemistry of DNA extraction, PCR amplification, and automated "Sanger" DNA sequencing.

6.    What are the most important parameters in designing oligonucleotide primers for PCR?

7.    From the accompanying data, construct a Restriction Map of the target DNA.
